“We assessed the effects of two REV-ERB agonists, SR9009 and SR9011, on obesity in diet-induced obese mice. In addition, we determined the impact of REV-ERB activation on circadian rhythm (CR) and insulin sensitivity. Both drugs reduced the weight of epididymal (epiWAT) and perirenal adipose tissue (pWAT) of the mice. At the molecular level, both drugs increased the activity of the enzymes responsible for fatty acid β-oxidation and triglyceride hydrolysis in liver and muscle. Activation of REV-ERB also induced lipid mobilization (i.e., increased circulating non-esterified fatty acids) in the mice. In addition, the CR of the mice was increased. Our findings suggest that REV-ERB activation could be a novel therapeutic strategy for obesity and type 2 diabetes mellitus.”
